Jonah Jameson turned to ESU electro-biologist Dr. Marla Madison to help construct his next Spider-Slayer. Suggesting that she's run out of challenges, he recruits her in his latest scheme to destroy Spider-Man. Together, the two come up with the Spider-Slayer Mark V. The new version is equipped with an impulse-beam, web-shears, grappling-claw, and strength enough to curl an iron bar. The Mark V is controlled with a psycho-cybernetic helmet.

Jameson Sends out this Spider-Slayer to follow Peter Parker reasoning that Parker will eventually lead him to Spider-Man. Peter picks up that he's being shadowed, and as Spider-Man, confronts the new Spider-Slayer. After taunting Jameson a bit, Spider-Man declines to battle the slayer and swings off instead.

Jameson's Spider-Slayer catches up again just as Spider-Man has been incapacitated by Will-O'-The-Wisp. The Spider-Slayer battles Will-O'-The-Wisp in the Rockefeller Plaza skating rink for the right to destroy Spider-Man. During the melee, Will-O'-The-Wisp gets mysteriously recalled, but by Spider-Man has recovered by that time. Spider-Man manages to defeat the Spider-Slayer by crushing it under the giant status of Prometheus that crowns skating rink. Dr. Madison vows to restructure the Spider-Slayer and make it more powerful than ever.
